Two PHGDH subunits oligomerize to convert 3-phosphoglycerate to phosphohydroxypyruvate in an NAD + -dependent reaction. WebThe first enzyme of this pathway, phosphoglycerate dehydrogenase (PHGDH) is overexpressed in many types of cancers such as breast, lung and melanoma. Inhibitors of PHGDH have been designed (NCT-503 and CBR-5884) , although they present brain cytotoxic effects and its effectivity depends on environmental serine availability.
